| I. Background | |

TREM2 是免疫球蛋白超家族的單次跨膜受體，主要表達于巨噬細胞、未成熟單核細胞 衍生的樹突狀細胞、破骨細胞和小膠質細胞上。TREM2 包含免疫球蛋白 V 型結構的胞外 域、跨膜域和沒有信號傳導基序的短胞內結構域。它通過 DNAX 激活蛋白 10 和 12 （DAP10 和 DAP12）發(fā)出信號。單個 TREM2 分子可以同時與 DAP10 和 DAP12 相互作用。 TREM2 與陰離子配體結合，例如磷脂、硫苷脂、細菌脂多糖(LPS) 和 DNA。 TREM2–DAP12–DAP10 信號傳導觸發(fā)蛋白質和脂質磷酸化級聯(lián)，導致 Ca2+ 動員、整合 素激活、細胞骨架重排、雷帕霉素的機制靶點(mTOR) 和絲裂原活化蛋白激酶(MAPK) 信號 傳導以及能量代謝的激活。 |
