| I. Background | |

TL1A 又稱 TNFSF15，是 TNF 超家族的成員，在單核細胞、巨噬細胞、樹突狀細胞、 T 細胞等免疫細胞和非免疫細胞（如滑膜成纖維細胞、內(nèi)皮細胞）中均有表達。TL1A 是一 種 Ⅱ 型跨膜蛋白，通過與 TNF 同源結(jié)構(gòu)域 (THD) 相互作用自組裝成穩(wěn)定的三聚體。它主 要以膜結(jié)合形式表達，并形成穩(wěn)定的三聚體。可溶性 TL1A (sTL1A) 是通過可變剪接或 TNF-α轉(zhuǎn)換酶 (TACE) 裂解產(chǎn)生的。
TL1A 競爭性地與死亡受體 3（DR3）或誘餌受體 3（DcR3）結(jié)合，為下游信號通路提 供刺激信號，進而調(diào)控效應(yīng)細胞的增殖、活化、凋亡及細胞因子、趨化因子的產(chǎn)生。 |
